SCOPE:: Sulforaphane is a naturally occurring isothiocynate compound found in cruciferous vegetables. Here we report the effect of sulforaphane on cardiac hypertrophy and propose an underlying mechanism. METHODS AND RESULTS:: Sulforaphane suppresses cardiomyocyte hypertrophy induced by hypertrophic stimuli in vitro and in vivo. Sulforaphane suppresses the expressions of fetal genes, including ANP, BNP, and β-MHC. We used an siRNA technique and ANP promoter with mutated GATA binding sites to demonstrate that sulforaphane mediates cardiac hypertrophy by modulating transcription factors GATA4/6. CONCLUSION:: These results suggest that sulforaphane has the potential to prevent cardiac hypertrophy by down-regulating GATA4/6 and MAPK signaling pathways. This article is protected by copyright.
